I need to link my text box to my button allowing me to disable it and enable it at will but I can't conect them trough code.

First, you need to create the button event.
Double click to the button on your form, then the VB will create a button event automatically.
Now, you can code.

I try to save the changed text of an editable div. I put some text on page load into the div and change it in the browser, but I can't save the changes because the innerHTML content doesn't change.

Make a little trick with JavaScript.
When the button is clicked, get the innerHTML of DIV and put into a asp:HiddenField

The problem is - I can't set the value of MyNewValue before the form is submitted to the remote address https://some-remote-site/Form.aspx.

You can do it client-side using jQuery. So when the user clicks submit, and before submitting the form, you can update your hidden input and then return true to proceed the submission of your form.
This is it could be done:

I am attempting to recapture some form values after they have been posted, but am having quite a bit of difficulty. My form has 2 fields:

Without seeing the rest of the .aspx page, there's nothing glaringly obviously wrong there, but:
are the TextBoxes within a form with runat="server"?
have you run a trace to see what the Form variables, if any, contain?
are you sure the page is being submitted, and not just refreshed?
have you tried a breakpoint to see whether the if condition evaluates to true (similar to above point)?
I would think you can add onclick= to the button that is submitting the form since the textboxes are running at server. After the equal sign if you press tab twice it will automatically generate an event in the code behind. Then you can do something like this in the code behind to save it in session.

Even though, I put the script in a function to change the color, it is still not doing it and why do I need to add the script tags to true if the function is already enclosed in script tags?
lblDisplayDate is in the Page Load where it is set to the current time on every page reload.
Change this line:

Also, this probably isn't doing what you want it to do anyway. Try adding:
OnClientClick="changecolor()"
to your <asp:Button /> tag.
Edit At this point I am pretty confused as to what exactly you are trying to accomplish...
If you just want to change the color of the label without posting back, change your <asp:Button /> to a normal <input type="button" /> with an onclick handler and call your script, like this:
<input type="button" ... onclick="changeColor();" />
If you want to change the color of the label while posting back, then just change the label's background in the code-behind like this:

like what Sean already stated: it is best to use OnClientClick in this case. OnClientClick would prevent a postback occuring - it is done dynamically using javascript. All u need to do is preference it to the javascript function.
I do not understand y u want to do a "RegisterStartupScript and RegisterClientScriptBlock" on colour change. These should be used when u want to add javascript from the code behind, but seeing that u have it written out already in the aspx page, its use is pointless.

It seems that “asp:Button” does not work inside a “mmenu nav “.
Nor a "asp:LinkButton" seems to fully perform.

Web forms most of the time change the name of the element and jquery needs the exact name to find your button. Add this property to your button and test again
ClientIDMode="Static"
The mmenu plugin grabs the NAV#login from the HTML and puts it back directly inside the BODY. Meaning, the input and button are no longer inside the FORM (and thus won't submit the form).
Basically, if you put the FORM inside the NAV#login all should work as expected.
